Description

Semax acetate is a BBB-penetrable adrenocorticotropic hormone-like peptide and can form stable complexes with Cu2+. Semax acetate is a synthetic peptide analog of Adrenocorticotropic hormone (ACTH) (4-10). Semax acetate has immunomodulatory, nootropic and neuroprotective activities. Semax acetate can be used in the research of central nervous system diseases such as Alzheimer's disease and cerebral ischemia[1][2][3][4][5].

In Vitro

Semax (25-100 μM, 48 h) acetate can reduce the levels of Aβ oligomers[1].
Semax (100 nM) acetate can increase the survival rate of cholinergic neurons in the basal forebrain and stimulate the activity of choline acetyltransferase in cultured basal forebrain tissue ex vivo[2].
Semax (10 μM, 0-4 h) acetate can increase the levels of BDNF and NGF in glial cells derived from the basal forebrain of newborn rats[3].

In Vivo

Semax (100 μg/kg; intraperitoneal injection; single dose) acetate has an ameliorating effect in a rat model of focal cerebral ischemia[4].
Semax (0.15 mg/kg; intraperitoneal injection; single dose) acetate can increase the concentration of 5-HIAA in the hypothalamus and striatum of mice[5].

Animal Model: Ischemia (pMCAO) rats[4]
Dosage: 100 μg/kg
Administration: Intraperitoneal injections (i.p.), performed 15 min, 1, 4 and 8 h after permanent middle cerebral artery occlusion (pMCAO).
Result: Affected the expression of genes related to the immune system. At 3 hours after pMCAO, it influenced the expression of genes regulating the activity of immune cells such as macrophages, neutrophils, and lymphocytes. At 24 hours after pMCAO, the effect on the immune response was significantly increased, with over 50% of the genes with altered expression involved in the immune response.
Affected the expression of genes related to the vascular system. At 3 hours after pMCAO, it influenced genes involved in vasculogenesis, hematopoiesis, and endothelial cell migration. At 24 hours after pMCAO, it affected the development of endothelial tissue, the migration of smooth muscle cells, and the activation of blood cells.
