Description

RW3 (MP196) is a small cationic antimicrobial hexapeptide. RW3 targets the bacterial cytoplasmic membrane and inhibits cellular respiration and cell wall synthesis. RW3 exhibits high bioactivity against Gram-positive bacteria such as Bacillus subtilis 168 (MIC of 2 µg/mL). RW3 causes shrinking of murine erythrocytes. RW3 can be used in antimicrobial and antifungal research[1].

In Vitro

RW3 (16 h) shows decent activity against Gram-positive bacteria like Bacillus subtilis 168 (MIC of 2 µg/mL) and some MRSA/VISA strains, but weak activity against Gram-negative bacteria such as Pseudomonas aeruginosa PA01 (MIC of >64 µg/mL)[1].
RW3 (0.09-200 µg/mL; 72 h) shows no significant cytotoxicity against NRK-52E rat kidney epithelial cells and CCRF-CEM human T-cell lymphoblast cells[1].
RW3 (10 ng-500 µg/mL; 1-24 h) does not induce NF-κB activation in RT4 bladder epithelial cells[1].
RW3 (250-500 µg/mL) causes shrinking of murine erythrocytes at 250 µg/mL and severe morphological changes plus lysis at 500 µg/mL, with 14% and 23% hemolysis respectively[1].

In Vivo

RW3 (5-25 mg/kg; i.v.) causes excitement and indisposition (recovered within 25 min) at 5-10 mg/kg, and hind limb paralysis plus death within minutes at 25 mg/kg in mice[1].
