Wnt3a Protein, Human is an activator of the Wnt/β-catenin pathway that inhibits GSK-3β, stabilizes β-catenin and promotes its nuclear translocation to activate FOXM1. Wnt3a Protein is mainly used in studies on neuroprotection in ischemic stroke (such as reducing cerebral infarction volume in MCAO models)[1][2]. Wnt3a Protein, Human (His) is a recombinant human Wnt3a protein expressed in E. coli with a 6His tag at the N-terminus.

Background

Wnt3a Protein is a glycosylated secretory glycoprotein, a classic member of the Wnt family. Its structure contains an N-terminal cysteine-rich domain that can specifically bind to Frizzled family receptors (such as Frz1). Wnt3a Protein is an endogenous agonist of the Wnt/β-catenin signaling pathway. It binds to Frz1 and the co-receptor LRP6, inhibits downstream GSK-3β phosphorylation, relieves β-catenin degradation and promotes its nuclear translocation, thereby activating the transcription of anti-apoptotic genes such as FOXM1, while downregulating apoptotic markers such as Cleaved Caspase-3. In addition, Wnt3a can enhance β-catenin stability by activating PIWIL1a, and synergistically regulate cell survival and differentiation. The biological activity of Wnt3a Protein also includes promoting the differentiation of human umbilical cord mesenchymal stem cells (hUCMSCs) into epidermal-like cells, which is achieved by upregulating CK19 expression and β-catenin signaling. Wnt3a Protein also interacts with phosphatidylinositol proteoglycans GPC3 and PKD1 through its extracellular domain, participates in normal embryonic mesoderm development, caudal somite formation and the morphogenesis of the developing neural tube, and mediates the self-renewal of intestinal crypt base stem cells in vitro. Wnt3a Protein is mainly used for neuroprotection of ischemic stroke (such as reducing the volume of cerebral infarction in MCAO models and improving neurological function) and directed differentiation induction of stem cells in skin tissue engineering, providing potential targets for stroke treatment and trauma repair[1][2].
