Background

WNK4 is a serine/threonine-protein kinase that serves as a key component of the WNK4-SPAK/OSR1 kinase cascade, playing a critical role in ion transport regulation in the distal nephron and blood pressure control (By similarity). Within this cascade, WNK4 phosphorylates and activates downstream kinases OXSR1/OSR1 and STK39/SPAK, which in turn modulate the activity of ion cotransporters such as SLC12A1/NKCC2 and SLC12A3/NCC. Functioning as a molecular switch, WNK4 balances renal salt reabsorption and potassium secretion by regulating transporters and channels including SLC12A3/NCC and KCNJ1/ROMK (By similarity). Independently of its kinase activity, WNK4 acts as a scaffold protein to negatively regulate membrane localization of various transporters/channels (e.g., CFTR, TRPV4) via clathrin-dependent endocytosis (By similarity). It also inhibits epithelial sodium channel (ENaC) activity through a kinase-independent mechanism (By similarity) and may phosphorylate NEDD4L.
