TIGIT Protein, Rat (122a.a, HEK293, Fc)

TIGIT is a kind of inhibitory immune checkpoint, which can exert immunosuppressive function by directly or indirectly inhibiting effector T cells and up-regulating Treg cells. TIGIT shows low affinity binding to NECTIN2 and NECTIN3. TIGIT Protein, Rat (122a.a, HEK293, Fc) is the recombinant rat-derived TIGIT protein, expressed by HEK293 , with C-hFc labeled tag. The total length of TIGIT Protein, Rat (122a.a, HEK293, Fc) is 122 a.a., with molecular weight of 48-55 kDa.

TIGIT is an inhibitory immune checkpoint. By binding with CD155 and other antibodies, Tigit can promote the depletion of NK cells and reduce the secretion of cytokines. It can also exert immunosuppressive function by directly or indirectly inhibiting effector T cells and up-regulating the role of Treg cells. TIGIT plays a synergistic inhibitory role with PD-1/PD-L1, TIM-3 and other co-inhibitory receptors. TIGIT protein plays a key role in immune regulation and binds to poliovirus receptor (PVR) with high affinity. This interaction results in increased IL10 secretion and decreased IL12B secretion, resulting in an immunosuppressive environment. TIGIT further plays an immunomodulatory role by inhibiting T cell activation and promoting the generation of mature immunomodulatory dendritic cells. TIGIT shows low affinity binding to NECTIN2 and NECTIN3^[1][2][3].

Immobilized Human Nectin-2 His at 5 μg/mL (100 μL/well) can bind Rat TIGIT Fc with a linear range of 0.039-5 μg/mL.

Lyophilized a 0.22 μm filtered solution of Tris with Glycine, Arginine and NaCl, pH 7.5. Normally trehalose is added as protectant before lyophilization.

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH₂O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
