RIIB/ACVR2B Protein, Rat (HEK293, His)

RIIB/ACVR2B Protein, Rat (HEK293, His) is the recombinant rat-derived RIIB/ACVR2B protein, expressed by HEK293, with C-His tag.

RIIB/ACVR2B is a transmembrane serine/threonine kinase acting as an activin type-2 receptor, which forms a receptor complex with activin type-1 serine/threonine kinase receptors (e.g., ACVR1, ACVR1B, or ACVR1c). It transduces activin signals from the cell surface to the cytoplasm, regulating diverse physiological and pathological processes such as neuronal differentiation/survival, hair follicle development/cycling, pituitary FSH production, wound healing, extracellular matrix synthesis, immunosuppression, and carcinogenesis. Activin may also function in ovarian follicular development via paracrine/autocrine mechanisms. Within the complex, type-2 receptors (e.g., ACVR2B) serve as primary receptors binding activin-A/INHBA, activin-B/INHBB, and inhibin-A/INHA-INHBA, while type-1 receptors (e.g., ACVR1B) act as downstream signal transducers. Activin binding activates the type-2 receptor's kinase, leading to phosphorylation and activation of type-1 receptors. Activated type-1 receptors phosphorylate SMAD2/SMAD3 at C-terminal serines, enabling their cytoplasmic association with SMAD4 and nuclear translocation to mediate transcriptional responses. Inhibitory SMAD7, recruited to ACVR1B via FKBP1A, blocks SMAD2/3-receptor interaction to terminate signaling (By similarity, inhibin-B antagonizes activin signaling by binding the receptor through the IGSF1 coreceptor).
