Background

RANKL/TNFSF11 is a cytokine that binds to TNFRSF11B/OPG and TNFRSF11A/RANK. It acts as an osteoclast differentiation and activation factor, enhances the ability of dendritic cells to stimulate naive T-cell proliferation, and may be a key regulator of T-cell–dendritic cell interactions and T-cell-dependent immune responses. It also likely contributes to increased bone resorption in humoral hypercalcemia of malignancy. By activating multiple signaling pathways in osteoclast precursor cells—particularly through inducing prolonged intracellular Ca 2+ oscillations that activate NFATC1 (leading to its nuclear translocation and induction of osteoclast-specific gene transcription)—RANKL/TNFSF11 promotes osteoclastogenesis. During osteoclast differentiation, it also triggers TMEM64- and ATP2A2-dependent CREB1 activation and mitochondrial ROS generation, which are essential for proper osteoclast formation (By similarity).
