Background

PTBP1 is a protein involved in pre-mRNA splicing and the regulation of alternative splicing events. It activates exon skipping of its own pre-mRNA during muscle cell differentiation and binds to the polypyrimidine tract of introns. When bound to two separate polypyrimidine tracts in the same pre-mRNA, it may promote RNA looping and facilitate U2 snRNP binding to pre-mRNA. PTBP1 cooperates with RAVER1 to modulate switching between mutually exclusive exons during TPM1 pre-mRNA maturation and represses the splicing of MAPT/Tau exon 10. Additionally, it binds to the polypyrimidine-rich controlling element (PCE) of CFTR, promoting exon 9 skipping and antagonizing TIA1's role in exon inclusion. In pyruvate kinase PKM splicing, PTBP1 repressively binds to a polypyrimidine tract flanking exon 9, inhibiting its inclusion and favoring exon 10 inclusion to produce the PKM M2 isoform. During picornavirus infection, PTBP1 binds to the viral internal ribosome entry site (IRES), stimulating IRES-mediated translation.
