Background

PRPF4B is a serine/threonine kinase involved in spliceosomal assembly, mitosis, and signaling regulation. It bridges chromatin-mediated transcriptional regulation with pre-mRNA splicing, interacting with and phosphorylating U4/U6-U5 tri-snRNP components PRPF6 and PRPF31 to facilitate spliceosome B complex formation. PRPF4B also regulates transcription and the spindle assembly checkpoint (SAC), associating with U5 snRNP and NCOR1 deacetylase complexes to coordinate splicing with chromatin remodeling. It likely phosphorylates SMARCA4 and NCOR1, as well as SRSF1. During mitosis, PRPF4B localizes to kinetochores and is essential for recruiting/maintaining checkpoint proteins (e.g., MAD1L1, MAD2L1). Additionally, it phosphorylates transcription factors ELK1, KLF13, and nuclear YAP1/WWTR1(TAZ), the latter triggering nuclear exclusion and modulating the Hippo pathway (implicated in tissue growth control).
