Background

PRPF19 is a ubiquitin-protein ligase that serves as a core component of several complexes primarily involved in pre-mRNA splicing and DNA repair. As part of the spliceosome, it facilitates assembly, remodeling, and activity, including mediating 'Lys-63'-linked polyubiquitination of the U4 spliceosomal protein PRPF3 to stabilize the U4/U5/U6 tri-snRNP complex. Recruited to RNA polymerase II CTD, it may couple transcription with splicing machinery. In DNA damage response (DDR), PRPF19 (within the PRP19-CDC5L complex) ubiquitinates RPA1/RPA2 at damage sites to activate the ATR pathway. It also contributes to DNA double-strand break (DSB) repair by recruiting SETMAR and potentially participates in DNA interstrand cross-link (ICL) repair via the PSO4 complex. PRPF19 may additionally mediate 'Lys-48'-linked polyubiquitination for proteasomal degradation. By similarity, it might play roles in lipid droplet biogenesis and neural differentiation through spliceosomal functions.
