Background

Protein phosphatase 1 regulatory subunit 1A is a potent inhibitor of protein phosphatase 1 (PP1). PPP1R1A plays an important role in the hormonal control of glycogen metabolism. Hormones that raise intracellular cAMP increase I-1 activity in many tissues. I-1 activation may enable cAMP to control proteins that are not directly phosphorylated by PKA. As intracellular calcium rises, I-1 is inactivated by calcineurin (or PP2B). Does not inhibit type 2 phosphatase. PPP1R1A may regulate activation of PI3K-Akt pathway, cell cycle, glycogen metabolism, and recruitment of M2 macrophages in liver cancer. PPP1R1A regulates GLR1R-mediated expansion of glucose-stimulated insulin secretion in beta cells. Inhibition of PKA/PPP1R1A/PP1 pathway has potential therapeutic value in the treatment of both primary and metastatic Ewing's sarcoma (ES)[1][2][3][4].
