Upon ligand activation, the receptor engages with promoter elements of target genes, regulating the peroxisomal beta-oxidation pathway of fatty acids and serving as a transcription activator for the acyl-CoA oxidase gene. Additionally, it plays a role in decreasing the expression of NPC1L1 upon ligand activation. PPARD forms a heterodimer with the retinoid X receptor and interacts with CRY1 and CRY2 in a ligand-dependent manner via its NR LBD domain.
