Background

PHLPP2 is a protein phosphatase involved in the regulation of Akt and PKC signaling pathways. It mediates dephosphorylation of the C-terminal hydrophobic motif in members of the AGC Ser/Thr protein kinase family, specifically targeting 'Ser-473' of AKT1, 'Ser-660' of PRKCB isoform beta-II, and 'Ser-657' of PRKCA. Akt regulates the balance between cell survival and apoptosis by modulating transcription factors that control pro- and anti-apoptotic genes. Dephosphorylation of Akt at 'Ser-473' by PHLPP2 promotes apoptosis and reduces cell proliferation. PHLPP2 also regulates the phosphorylation of AKT3 and dephosphorylates STK4 at 'Thr-387', leading to STK4 activation and apoptosis. Additionally, it dephosphorylates RPS6KB1, influencing cap-dependent translation. PHLPP2 inhibits cancer cell proliferation and may function as a tumor suppressor. Dephosphorylation of PRKCA and PRKCB by PHLPP2 destabilizes and degrades these proteins. Furthermore, PHLPP2 dephosphorylates RAF1, suppressing its kinase activity.
