NUAK1 is a serine/threonine-protein kinase involved in diverse processes including cell adhesion, regulation of cell ploidy and senescence, cell proliferation, and tumor progression. It phosphorylates substrates such as ATM, CASP6, LATS1, PPP1R12A, and p53/TP53. Key functions include: 1) regulating cellular senescence and ploidy by phosphorylating LATS1 at Ser-464 to control its stability; 2) modulating cell adhesion via phosphorylation of the PPP1R12A subunit of myosin PP1 complex, which recruits 14-3-3 proteins to inhibit MLC2 dephosphorylation; 3) potential role in DNA damage response by phosphorylating p53 at Ser-15/Ser-392 and participating in p53-mediated CDKN1A/WAF1 promoter activation; 4) acting as a tumor malignancy factor under AKT1 regulation to promote invasion/metastasis; 5) suppressing Fas-induced apoptosis through CASP6 phosphorylation; 6) cooperating with STK11 to phosphorylate CDKN1A upon UV radiation, facilitating its degradation for efficient DNA repair. by similar: Its mechanisms overlap with LATS1 stabilization, PP1 complex inhibition, and p53 pathway activation.
