Background

NT5C2 is a broad specificity cytosolic 5'-nucleotidase that primarily catalyzes the dephosphorylation of 6-hydroxypurine nucleoside 5'-monophosphates. It also exhibits phosphotransferase activity, transferring phosphate groups from donor nucleoside monophosphates to acceptor nucleosides (with preference for inosine, deoxyinosine, and guanosine). The enzyme shows highest activity toward IMP and GMP, followed by dIMP, dGMP, and XMP; while it can catalyze phosphate transfer from pyrimidine monophosphates, the efficiency is lower. These activities enable NT5C2 to regulate cellular purine nucleoside/nucleotide pools. by similar: Its phosphotransferase activity is more pronounced for inosine, deoxyinosine, and guanosine, with relatively reduced efficiency for pyrimidine monophosphates.
