NA/Neuraminidase Protein, Influenza A H1N1 (HEK293)

Cat. No.: HY-P73231
Handling Instructions Technical Support

NA/Neuraminidase Protein is a receptor-destroying enzyme that cleaves terminal sialic acids from cellular receptors, potentially facilitating viral invasion of the upper airways by targeting sialic acid moieties on airway epithelial cell mucin. NA plays a pivotal role in viral propagation by catalyzing the removal of terminal sialic acid residues from both viral and cellular glycoconjugates. Moreover, the sialidase activity in late endosome/lysosome traffic appears to enhance virus replication. This product is an active extract, not a recombinant NA protein. It originates from cell lysates collected after expressing a DNA sequence encoding the Influenza A virus neuraminidase.

Description

NA/Neuraminidase Protein is a receptor-destroying enzyme that cleaves terminal sialic acids from cellular receptors, potentially facilitating viral invasion of the upper airways by targeting sialic acid moieties on airway epithelial cell mucin. It originates from cell lysates collected after expressing a DNA sequence encoding the Influenza A virus neuraminidase.

Background

Neuraminidase (NA) plays a pivotal role in viral propagation by catalyzing the removal of terminal sialic acid residues from both viral and cellular glycoconjugates. Specifically, during virus budding, NA cleaves off terminal sialic acids from the glycosylated hemagglutinin (HA), facilitating the release of viral particles and enabling efficient virus spread through the circulation. By preventing self-aggregation and ensuring the removal of sialic acids from the cell surface, NA allows the progeny virus to disseminate efficiently from cell to cell, thereby avoiding limitations to a single round of replication. Described as a receptor-destroying enzyme, NA cleaves terminal sialic acids from cellular receptors, potentially facilitating viral invasion of the upper airways by targeting sialic acid moieties on airway epithelial cell mucin. Its association with lipid rafts during intracellular transport, and its potential raft-independent effect on budding, highlight the multifaceted role of NA in determining host range restriction, replication, and virulence. Moreover, the sialidase activity in late endosome/lysosome traffic appears to enhance virus replication[1][2][3].

Biological Activity

Measured by its ability to cleave a fluorogenic substrate , 2’-(4- Methylumbelliferyl)-α-D-N-acetylneuraminic acid. One unit is defined as the amount of enzyme required to cleave 1 nmole of 2'-(4-Methylumbelliferyl)-α-D-N-acetylneuraminic acid per minute at pH 7.5 at 37℃.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

  • Reconstitution Calculator

  • Dilution Calculator

  • Specific Activity Calculator

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
