Description

MUC15 is a membrane-bound mucin. The human MUC15 protein contains an extracelluar domain, a small transmembrane domain, and a highly conserved cytoplasmic tail. MUC15 overexpression suppresses trophoblast-like cell invasion[1][2]. Mucin-15/MUC15 Protein, Cynomolgus (HEK293, Fc) is the recombinant cynomolgus-derived Mucin-15/MUC15 protein, expressed by HEK293 , with C-hFc labeled tag.

Background

MUC15 (mucin) was characterized because of its association with the bovine milk fat globule membrane (MFGM). It was shown to be a 33.3-kDa (307 AA) protein comprising an extracellular mucin domain rich in Ser, Thr, and Pro residues, a type 1 membrane-spanning domain, and a short cytoplasmic tail. In SDS-PAGE, bovine MUC15 migrates as a 115- to 130-kDa protein, which reflects its massive glycosylation (both N- and O-glycans). The corresponding cDNA of human MUC15 encodes an unprocessed protein of 334 AA showing 67% sequence similarity with the bovine counterpart[1].
The membrane-bound mucins, which are type I membrane proteins, include MUC1, MUC3, MUC4, MUC12, MUC13, MUC15, MUC16, MUC17, and MUC20. Overexpression of MUC15 substantially decreased matrigel invasion of JAR and JEG-3 cells by 87.5 ± 1.1 and 83.8 ± 5.7%, respectively. The low levels of MUC15 expression may be critical for trophoblast invasion in early placental development, whereas high levels of MUC15 expression may provide a suppressive signal for trophoblast invasion at the end of gestation[2].

Formulation

Lyophilized a 0.22 μm filtered solution of 50 mM Tris, 100 mM Glycine, 25 mM Arginine, 150 mM NaCl, pH 7.5. Normally trehalose is added as protectant before lyophilization.
Note: For SPR assay, please replace the buffer. Primary amine components (e.g., Tris, imidazole) can affect protein-coupled chips.

Reconstitution

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH2O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
