MAdCAM1 Protein is an important cell adhesion molecule and a member of the immunoglobulin superfamily. MAdCAM1 Protein plays a key role in immune system cell migration and mucosal immune responses. MAdCAM1 Protein, Human (HEK293, His) is a recombinant MAdCAM1 protein expressed by HEK293, with a C-6*His tag and glycosylation modification[1].

Background

MAdCAM1 is a transmembrane glycoprotein expressed by endothelial cells, primarily located in the vascular endothelium of gut-associated lymphoid tissues and blood vessels at chronic inflammatory sites. MAdCAM1 mediates the homing of lymphocytes to intestinal mucosal tissues and organs such as the liver under inflammatory conditions by binding to the integrin α4β7 on the surface of lymphocytes. MAdCAM1 is upregulated during intestinal inflammation or infection to promote lymphocyte infiltration and enhance local immune defense, and its abnormal expression is closely associated with inflammatory bowel disease[1].

In Vitro

PF-00547659 binds to MAdCAM1 Protein, Human and inhibits the adhesion of α4β7-expressing cells to human and cynomolgus MAdCAM in vitro[2].

Biological Activity

1.Measured by the ability of the immobilized protein to support the adhesion of Jurkat human T-lymphocyte leukemia cells in the presence of 1 mM MnCl2. The ED50 for this effect is 1.987 μg/mL, corresponding to a specific activity is 503.2 units/mg.
2.Measured by the ability of the immobilized protein to support the adhesion of Jurkat human T-lymphocyte leukemia cells. When 5×104 cells/well are added to Human MAdCAM-1 coated plates (2.5 μg/mL with 100 µL/well), approximately 52.575%-62.728% will adhere after 1 hour incubation at 37 °C in the presence of 1 mM MnCl2.

  • Measured by the ability of the immobilized protein to support the adhesion of Jurkat human T-lymphocyte leukemia cells in the presence of 1 mM MnCl2. The ED50 for this effect is 1.987 μg/mL, corresponding to a specific activity is 503.2 units/mg.

Reconstitution
