LINGO-1 serves as a functional component within the Nogo receptor signaling complex (RTN4R/NGFR), playing a crucial role in RhoA activation and contributing to the inhibition of axonal regeneration caused by myelin-associated factors. This protein is also a key negative regulator of oligodendrocyte differentiation and axonal myelination. In conjunction with RTN4 and RTN4R, LINGO-1 participates in the regulation of neuronal precursor cell motility during cortical development. It forms a homotetramer and engages in a ternary complex with RTN4R/NGFR and RTN4R/TNFRSF19. LINGO-1 further interacts with NGRF and MYT1L, highlighting its multifaceted involvement in neurodevelopmental processes and myelin-associated inhibition.
