Background

The Integrin alpha-IIb/beta-3 protein is a multifaceted receptor with broad ligand recognition capabilities, including binding to fibronectin, fibrinogen, plasminogen, prothrombin, thrombospondin, and vitronectin through the R-G-D sequence. Specifically, it recognizes the H-H-L-G-G-G-A-K-Q-A-G-D-V sequence in the fibrinogen gamma chain. Upon activation, Integrin alpha-IIb/beta-3 facilitates platelet-platelet interactions by binding soluble fibrinogen, leading to rapid platelet aggregation that forms a physical plug on the ruptured endothelial cell surface. Structurally, it forms a heterodimer, with the alpha subunit consisting of a heavy and a light chain linked by a disulfide bond, and the alpha-IIb subunit associates with beta-3. Additionally, Integrin alpha-IIb/beta-3 directly interacts with RNF181, and its cytoplasmic tail region interacts with CIB1, CIB2, CIB3, and CIB4 in a calcium and magnesium-dependent manner. Furthermore, its interaction with PPIA/CYPA is ROS and PPIase activity-dependent and is increased in the presence of thrombin. This diverse range of interactions highlights the integral role of Integrin alpha-IIb/beta-3 in platelet function and hemostasis.
