Background

IL-6 is a multifunctional cytokine secreted by immune cells (such as T cells, B cells, and macrophages) and non-immune cells (such as fibroblasts, endothelial cells, and hepatocytes). IL-6 transmits signals by binding to its receptor. As a cytokine rapidly and transiently produced in response to infection and tissue damage, IL-6 participates in host defense by stimulating acute-phase responses, hematopoiesis, and immune reactions. Its dysregulation can trigger chronic inflammation and autoimmune diseases. In the initial stage of inflammation, IL-6 is transported via blood to the liver to induce the synthesis of multiple acute-phase proteins. It also participates in regulating serum iron and zinc levels and platelet release. In immune responses, IL-6 promotes Th17 differentiation, inhibits Treg differentiation, and affects immune tolerance[1][2][3].

In Vitro

IL-6 Protein (Mouse; 25 ng/mL; 24 h) can promote the production of inflammatory factors MCP-1, MMP-3, CCL5, CXCL12 and the expression of hypertrophic markers COL10A1 and RUNX2 in primary mouse chondrocytes. The mechanism involves the activation of the JAK2/STAT3 pathway[3].

In Vivo

IL-6 Protein (Mouse; 0.3 μg/kg; intra-articular injection; single dose) can cause spinal hyperexcitability in rats[3].

Biological Activity

1.The ED50 is <0.02 ng/mL as measured by murine 7TD1 cells, corresponding to a specific activity of >5 × 107 units/mg.
2. Measured by its binding ability in a functional ELISA. When Recombinant Mouse IL-6 Protein is immobilized at 5 μg/mL (100 µL/well) can bind Biotinylated Recombinant Mouse IL-6 R alpha. The ED50 for this effect is 20-156 ng/mL.

  • Immobilized Mouse IL-6, His Tag at 5 μg/mL (100 μL/well) can bind Biotinylated Mouse IL-6 R alpha that produces 50% of the optimal binding response is found to be approximately 117.5 ng/mL.

Reconstitution

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH2O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.

    Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
