Background

IGF system has been demonstrated to regulate the growth of numerous tissues, such as neural tissue, lymphoid tissue, reproductive tissue, smooth muscle, endothelium, and bone. IGF-I/IGF-1, a key growth factor involved in cell growth, differentiation, survival, and cell cycle regulation, is secreted by mature osteoblasts, stored in the bone matrix, and released during bone resorption. The mitogenic effect of IGF-I/IGF-1 is mediated through its binding to IGF plasma membrane receptors. IGF-I/IGF-1 can stimulate osteoblast proliferation and differentiation, and also promote neuronal survival[1][2][3][4].

In Vitro

IGF-I/IGF-1 protein (Human; 50 ng/mL; 48 h) can activate the PI3K/Akt/mTOR signaling pathway and alleviate the apoptosis induced by 5-Heptadecylresorcinol (AR-C17; HY-N2673) in MCF-7 cells[3].

In Vivo

IGF-I/IGF-1 Protein (Human; 0.3-10 μg/day; intraventricular infusion; 7 days) exerts a neuroprotective effect in a mouse model of traumatic brain injury, promoting hippocampal neurogenesis and improving motor and cognitive functions[4].

Biological Activity

1.ED50 < 5 ng/mL, measured by a cell proliferation assay using FDC-P1 cells, corresponding to a specific activity of > 2.0×105 units/mg.
2. Measured in cell proliferation assay using MCF-7 human breast cancer cells. The ED50 for this effect is ≤65 ng/mL.

  • Measured in a serum-free cell proliferation assay using MCF-7 human breast cancer cells. The ED50 for this effect is 1.392 ng/mL. corresponding to a specific activity is 7.189×105 units/mg.

Reconstitution

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH2O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
