Background

GBP4 is an interferon (IFN)-inducible GTPase with crucial roles in innate immunity against a broad spectrum of bacterial, viral, and protozoan pathogens. It efficiently hydrolyzes GTP, predominantly yielding GDP. Upon infection, GBP4 is recruited to pathogen-containing vacuoles or vacuole-escaped bacteria, acting as a positive regulator of inflammasome assembly. This occurs by promoting the release of inflammasome ligands from bacteria. GBP4 facilitates pathogen-containing vacuole lysis, leading to the release of pathogens into the cytosol. In the cytosol, it promotes the recruitment of proteins, such as Gm12250/Irgb10, which mediate bacterial cytolysis. This liberation of ligands, including lipopolysaccharide (LPS) and double-stranded DNA (dsDNA), activates inflammasomes, such as the non-canonical CASP4/CASP11 inflammasome and AIM2 inflammasome, respectively. Additionally, GBP4 may play a role in erythroid differentiation.
