Background

GBA/glucocerebrosidase is a glucosylceramidase that catalyzes the hydrolysis of glucosylceramides/GlcCers (e.g., beta-D-glucosyl-(1<->1')-N-acylsphing-4-enine) into free ceramides (e.g., N-acylsphing-4-enine) and glucose within the lysosomal compartment. It plays a central role in complex lipid degradation and cellular membrane turnover and participates in the PKC-activated salvage pathway of ceramide formation through ceramide production. Additionally, GBA catalyzes cholesterol glucosylation via a transglucosylation reaction, transferring glucose from GlcCer to cholesterol, with GlcCers containing mono-unsaturated fatty acids (e.g., beta-D-glucosyl-N-(9Z-octadecenoyl)-sphing-4-enine) being preferred glucose donors over those with saturated fatty acids (e.g., beta-D-glucosyl-N-octadecanoyl-sphing-4-enine). Under specific conditions, GBA may reverse the reaction, transferring glucose from cholesteryl 3-beta-D-glucoside to ceramide, or hydrolyze cholesteryl 3-beta-D-glucoside to produce glucose and cholesterol. GBA also hydrolyzes galactosylceramides/GalCers (e.g., beta-D-galactosyl-(1<->1')-N-acylsphing-4-enine) and mediates galactose transfer between GalCers and cholesterol in vitro, albeit with lower activity than with GlcCers. In contrast, xylosylceramide/XylCer (e.g., beta-D-xyosyl-(1<->1')-N-acylsphing-4-enine) is a poor substrate for hydrolysis but serves as an efficient xylose donor for transxylosylation to form cholesteryl 3-beta-D-xyloside.
