Background

Fas, also known as CD95, serves as a receptor for TNFSF6/FASLG, initiating a cascade of apoptotic events crucial for programmed cell death. The adapter molecule FADD facilitates the recruitment of caspase-8 to the activated receptor, forming the death-inducing signaling complex (DISC) and initiating the cascade of caspases that mediate apoptosis. FAS-mediated apoptosis is implicated in peripheral tolerance induction and the antigen-stimulated suicide of mature T-cells. Additionally, secreted isoforms 2 to 6 of Fas have been identified, which function to block apoptosis in vitro. Fas engages in diverse protein interactions, including DAXX, HIPK3, RIPK1, FAIM2, BABAM2, FEM1B, FADD, NOL3, and CALM, forming a complex network that regulates cell death signaling. The interaction with various molecules highlights the multifaceted roles of Fas in cellular processes, emphasizing its significance in immune regulation and apoptosis modulation.
