EGLN1 Protein, Human (His) is the recombinant human-derived EGLN1, expressed by E. coli , with His labeled tag.

Background

EGLN1 is a cellular oxygen sensor that catalyzes the post-translational formation of 4-hydroxyproline in hypoxia-inducible factor (HIF) alpha proteins under normoxic conditions. It hydroxylates specific proline residues in each oxygen-dependent degradation domain (ODD) of HIF1A (N-terminal NODD and C-terminal CODD) and similarly acts on HIF2A, with a preference for the CODD site in both HIF1A and HIF1B. Hydroxylated HIFs are targeted for proteasomal degradation via the von Hippel-Lindau ubiquitination complex. Under hypoxia, attenuated hydroxylation allows HIFs to escape degradation, translocate to the nucleus, heterodimerize with HIF1B, and upregulate hypoxia-inducible genes. As the most critical isozyme under normoxia, EGLN1 regulates HIF1 stability and participates in hypoxia-influenced processes like angiogenesis in retinal and cardiac functions. Target proteins are preferentially recognized through a LXXLAP motif.
