Background

DNMT1 is a DNA methyltransferase that primarily methylates CpG residues, with a preference for hemimethylated DNA. During the S phase, it associates with DNA replication sites to maintain methylation patterns in newly synthesized strands, which is critical for epigenetic inheritance. In the G2 and M phases, DNMT1 binds to chromatin independently of replication to sustain DNA methylation. It plays a key role in preserving methylation patterns established during development and coordinates with histone methylation. DNMT1 mediates transcriptional repression by directly binding to HDAC2. In collaboration with DNMT3B and through the recruitment of CTCFL/BORIS, it participates in activating BAG1 gene expression by modulating dimethylation of histone H3 at H3K4 and H3K9 in the promoter region. In colorectal cancer (CRC) cells, DNMT1 likely forms a corepressor complex involved in KRAS-activated promoter hypermethylation and transcriptional silencing of tumor suppressor genes (TSGs) or other tumor-related genes. Additionally, DNMT1 maintains a transcriptionally repressive state of genes in undifferentiated embryonic stem cells (ESCs) and binds to promoter regions of TSGs, leading to their silencing and promoting tumor growth.
