Background

CERS6 is a ceramide synthase that catalyzes the transfer of the acyl chain from acyl-CoA to a sphingoid base, exhibiting high selectivity for palmitoyl-CoA (hexadecanoyl-CoA; C16:0-CoA). It can also utilize other acyl donors, though with reduced efficiency (By similarity). In de novo synthesis and salvage pathways, CERS6 N-acylates sphinganine and sphingosine bases to produce dihydroceramides and ceramides, respectively. Ceramides generated by CERS6 are involved in inflammatory responses (By similarity) and regulate metabolism and hepatic lipid accumulation (By similarity). Under high-fat diet conditions, CERS6-derived palmitoyl- (C16:0-) ceramides bind to the mitochondrial fission factor MFF, promoting mitochondrial fragmentation and contributing to obesity development (By similarity).
