Background

Complement component C7 is an integral part of the membrane attack complex (MAC), a crucial effector in the innate and adaptive immune responses. The MAC plays a pivotal role by forming pores in the plasma membrane of target cells, leading to cell lysis and destruction. C7 functions as a membrane anchor in this process, and it can exist as a monomer or dimer, with the potential to form multimeric rosettes when part of the C5b-7 complex. MAC assembly begins with the proteolytic cleavage of C5 into C5a and C5b, and subsequently, C5b binds sequentially to C6, C7, C8, and multiple copies of the pore-forming subunit C9. This orchestrated sequence highlights the essential role of C7 in the assembly of MAC, contributing to the immune system's ability to eliminate target cells.
