ATG7 Protein, Human (sf9) is the recombinant human-derived ATG7, expressed by sf9 insect cells, with tag free labeled tag.

Background

ATG7 is an E1-like activating enzyme involved in the two ubiquitin-like systems required for cytoplasm to vacuole transport (Cvt) and autophagy. It activates ATG12 for conjugation with ATG5 and the ATG8 family proteins for conjugation with phosphatidylethanolamine. Both systems are essential for ATG8 association with Cvt vesicles and autophagosome membranes. ATG7 is required for autophagic death induced by caspase-8 inhibition and facilitates LC3-I lipidation with phosphatidylethanolamine to form LC3-II, which localizes to autophagosomal membranes. Additionally, ATG7 regulates mitochondrial quantity and quality through mitophagy, maintaining basal levels to meet cellular energy demands and prevent excessive ROS production. It modulates p53/TP53 activity to control cell cycle and survival during metabolic stress. ATG7 also plays a key role in maintaining axonal homeostasis, preventing axonal degeneration, sustaining hematopoietic stem cells, forming Paneth cell granules, and promoting adipose differentiation. Furthermore, it regulates the liver clock and glucose metabolism by mediating time-dependent autophagic degradation of CRY1 (clock repressor) (By similarity).
