5HT2A is a G-protein coupled receptor primarily activated by 5-hydroxytryptamine (serotonin) and also serves as a receptor for various drugs and psychoactive substances, including mescaline, psilocybin, DOI, and LSD. Ligand binding triggers signaling via G(q)/G(11) G alpha proteins, activating phospholipase C-beta to release second messengers DAG and IP3, which modulate phosphatidylinositol 3-kinase activity and promote intracellular calcium release. Beta-arrestin family members inhibit G protein signaling while mediating alternative pathway activation. 5HT2A influences neural activity, perception, cognition, and mood, and plays a role in behavioral regulation (e.g., anxiety responses and psychoactive substance reactions). Additionally, it is involved in intestinal smooth muscle contraction (by similarity) and may contribute to arterial vasoconstriction (by similarity).
