Abstract

Dendrobium officinale Kimura & Migo polysaccharide (DOP) is one of the main bioactive components of traditional Chinese medicine. Although DOP can encourage the growth of blood vessels surrounding hair follicles, it is unknown if it can ameliorate skin photoaging by stimulating angiogenesis. This study used publicly available data sets. Analysis of GSE38308 and GSE274955 data sets showed impaired angiogenesis in photoaged skin. In the ultraviolet (UV) radiation-induced photoaging model of Kunming mice, local DOP administration was found to enhance skin elasticity, increase Collagen levels, promote angiogenesis, and demonstrate therapeutic potential for ameliorating photoaging in mice. In the UVB-induced photoaging model of human umbilical vein endothelial cells (HUVEC), UVB radiation induced cellular senescence and reduced the tube-forming capacity of HUVECs, whereas DOP treatment was found to enhance angiogenesis by activating the kinase insert domain receptor (KDR)/phosphatidylinositol 3-kinase (PI3K)-protein kinase B (Akt) signaling pathway, thereby ameliorating photoaging. These findings provide a novel mechanism for the amelioration of skin photoaging by DOP, highlighting its potential as a therapeutic agent for treating skin photoaging.
