Ladiratuzumab (hLIV22) is a humanized monoclonal antibody against zinc transporter LIV-1/ZIP6. Ladiratuzumab is conjugated to MMAE (HY-15162) via a cleavable dipeptide linker to synthesize an antibody-drug conjugate (ADC) Ladiratuzumab vedotin (HY-P99683). Ladiratuzumab vedotin selectively targets LIV-1 protein overexpressed on the surface of tumor cells, enters cells through antibody-mediated receptor endocytosis, releases MMAE to inhibit microtubule polymerization, and kills adjacent tumor cells with a bystander effect. Ladiratuzumab can be used in the study of solid tumors such as metastatic triple-negative breast cancer (mTNBC).

Ladiratuzumab (hLIV22) is a humanized monoclonal antibody against zinc transporter LIV-1/ZIP6. Ladiratuzumab ADC molecule, Ladiratuzumab vedotin (SGN-LIV1A), induces microtubule network disruption and inhibited cell growth in MCF-7 and BR0555 breast cancer cell line experiments, showing antigen-specific cytotoxicity[1].
LIV-1 is a transmembrane protein and an estrogen-induced gene in breast cancer cells. Ladiratuzumab vedotin inhibits cell proliferation in LIV-1-positive breast cancer cells in vitro in a dose-dependent manner and has anti-tumor activity targeting LIV-1[1][2].

Biological Activity
  • Immobilized LIV-1/SLC39A6 Protein, Human (HEK293, His, HY-P78749) can bind Ladiratuzumab. The EC50 for this effect is 8.853 ng/mL.
  • Flow cytometric analysis of 1X106 MCF-7 cells labeling Ladiratuzumab (HY-P99682, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Fluor 488-conjugated AffiniPure Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa (HY-P99001, blue) was used as the isotype control.
