HSP70-IN-7 is a nonselective HSP70 inhibitor with Kds of 4.71, 2.16 and 2.93μM against HSP70, GRP75, and GRP78. HSP70-IN-7 exhibits selective toxicity against breast cancer cells, induces cell apoptosis and effectively suppresses the properties of breast cancer stem cells (BCSCs). HSP70-IN-7 (compound 5) (48 h) inhibits breast cancer cells proliferation with IC50 values of 0.793, 0.297, 0.863 and 0.6 μM against MDA-MB-231, BT549, MCF7 and CAMA1 cells[1].
HSP70-IN-7 (1 μM, 48 h) induces apoptosis in stable clones from NS/GRP75 shRNA-transfected BT549 cells[1].
HSP70-IN-7 (0.5-1 μM, 10-14 d) inhibits the colony formation and mammosphere formation of MDA-MB-231 cells[1].
HSP70-IN-7 (1 μM, 0-1 h) directly acts on mitochondrial GRP75, destroying its PPIs, thereby leading to mitochondrial membrane permeabilization and Cyt c release in BT549 cells[1].
