  3. Bafilomycin B1

Bafilomycin B1 is a macrolide antibiotic that can be isolated from mycelium of Streptomyces gresuis. Bafilomycin B1 is neuroprotective against Chloroquine (HY-17589A)-induced death. Bafilomycin B1 is a structural analog of Bafilomycin A1 and has the ability to inhibit V-ATPase. Bafilomycin B1 inhibits significantly the oxidized low density lipoprotein (LDL)-induced accumulation of lipid droplets.

In Vitro

Bafilomycin B1 (Compound BafB1) (1 nM, 24 h) significantly inhibits the chloroquine-induced death of cerebellar granule neurons[1].
Bafilomycin B1 (Compound BafB1) (0.1-10 nM, 48 h) does not induce SH-SY5Y cell death or apoptosis at concentration ≤ 1 nM and significantly attenuates chloroquine-induced cell death with the addition of 50 μM chloroquine[2].
Bafilomycin B1 (5-10 nM) significantly inhibits the [14C]oleate incorporation into cholesteryl esters in J774 cells incubated with [14C]oleate in the presence of oxidized LDL for 3 h[3].
Bafilomycin B1 (10-40 nM) significantly inhibits the surface binding, intracellular accumulation and degradation of oxidized 125I-LDL in J774 cells incubated with oxidized 125I-LDL at 37 ℃[3].
Bafilomycin B1 (50 nM, 20 min) inhibits 74% of the degradation of the internalized oxidized 125I-LDL in J774 cells, suggesting Bafilomycin B1 inhibits the endosomal and lysosomal functions[3].
Bafilomycin B1 (50 nM, 60 min) causes a reduction of the cell surface receptors for oxidized LDL when incubated with oxidized 125I-LDL at 37 ℃[3].

In Vivo

Bafilomycin B1 (Compound BafB1) (50-150 μg/mL, 24 h) shows significant protection against α-synuclein-induced dopaminergic degeneration in isogenic worms over-expressing human WT α-synuclein in dopamine neurons[2].

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
