Background

XBP1 functions as a transcription factor during endoplasmic reticulum (ER) stress by regulating the unfolded protein response (UPR). It is essential for cardiac myogenesis and hepatogenesis during embryonic development and participates in the development of secretory tissues such as the exocrine pancreas and salivary gland (By similarity). XBP1 is involved in the terminal differentiation of B lymphocytes into plasma cells and immunoglobulin production. It modulates the cellular response to ER stress in a PIK3R-dependent manner and binds to the cis-acting X box in the promoter regions of major histocompatibility complex class II genes. During embryonic development, XBP1 contributes to VEGF-induced endothelial cell (EC) proliferation and retinal blood vessel formation, as well as angiogenesis in adult tissues under ischemic conditions. Additionally, XBP1 serves as a major regulator of the UPR in obesity-induced insulin resistance and type 2 diabetes, aiding in obesity management and diabetes prevention (By similarity). XBP1 undergoes unconventional cytoplasmic splicing of its own mRNA, triggered by the ER transmembrane endoribonuclease ERN1. In ECs, XBP1 associates with KDR to promote IRE1-mediated production of XBP1 mRNA isoform 2 in a VEGF-dependent manner, driving EC proliferation and angiogenesis. XBP1 also acts as a negative feedback regulator of XBP1 isoform 2 protein levels via proteasome-mediated degradation, preventing constitutive activation of the ER stress response pathway (By similarity). Together with HDAC3, XBP1 activates NFE2L2-mediated HMOX1 transcription factor gene expression through a PI(3)K/mTORC2/Akt-dependent signaling pathway, supporting EC survival under disturbed flow/oxidative stress. XBP1 binds to the ER stress response element (ERSE), CRE-like sequences, Tax-responsive element (TRE), and TPA response elements (TRE).
