WDR79 is an RNA chaperone that plays a crucial role in telomere maintenance and RNA localization to Cajal bodies. It specifically recognizes and binds the Cajal body box (CAB box) found in small Cajal body RNAs (scaRNAs) and the telomerase RNA template component (TERC). As an essential component of the telomerase holoenzyme complex, which is vital for telomere elongation in most eukaryotes, WDR79 stimulates the complex's catalytic activity by binding the CAB box of TERC RNA and controlling the folding of its CR4/CR5 region. It also regulates the localization of the telomerase holoenzyme complex to Cajal bodies and facilitates TERC delivery to telomeres during S phase. Beyond telomere maintenance, WDR79 contributes to Cajal body formation by mediating scaRNA localization. In DNA damage repair, WDR79 is phosphorylated by ATM and relocalizes to DNA double-strand breaks, recruiting the ubiquitin ligase RNF8 to promote both homologous recombination (HR) and non-homologous end joining (NHEJ).
