VGluT1 Antibody (YA012)

VGluT1 Antibody (YA012) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to VGluT1.

VGluT1 is a multifunctional transporter that transports L-glutamate as well as ions such as chloride, proton, potassium, sodium, and phosphate. At the synaptic vesicle membrane, it primarily acts as a uniporter, preferentially transporting L-glutamate and phosphate from the cytoplasm into synaptic vesicles at presynaptic nerve terminals of excitatory neurons (By similarity). This L-glutamate or phosphate uniporter activity is electrogenic and driven by the proton electrochemical gradient, mainly the electrical gradient established by vacuolar H(+)-ATPase across the synaptic vesicle membrane (By similarity). Additionally, VGluT1 functions as a chloride channel, enabling chloride permeation through the synaptic vesicle membrane, which affects the proton electrochemical gradient and promotes vesicle acidification (By similarity). It may also act as a K(+)/H(+) antiporter to maintain the electrical gradient while reducing the chemical gradient, thereby sustaining vesicular glutamate uptake (By similarity). This vesicular K(+)/H(+) antiport activity is electroneutral (By similarity). At the plasma membrane, following exocytosis, VGluT1 operates as a Na(+) and phosphate symporter, transporting these from the extracellular space into the cytoplasm to regulate synaptic phosphate homeostasis (PubMed:10820226). The symporter activity is driven by an inside-negative membrane potential and is electrogenic (By similarity). VGluT1 is essential for synaptic signaling of visual-evoked responses from photoreceptors (By similarity).

  Western blot analysis of extracts from Neuro-2a(lane 2(20ug) and Neuro-2a(lane 3(40ug) using VGluT1 Antibody (HY-P80957) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P83730, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
