Background

KAP1 is a nuclear corepressor for KRAB domain-containing zinc finger proteins (KRAB-ZFPs), mediating gene silencing by recruiting the NuRD complex subunit CHD3 and histone methyltransferase SETDB1 (which catalyzes H3K9me) to KRAB target gene promoters. It enhances transcriptional repression by coordinating increased H3K9me, decreased H3K9ac/H3K14ac acetylation, and HP1 protein localization, with SETDB1 recruitment inducing heterochromatinization. KAP1 also acts as a coactivator for CEBPB and NR3C1 in ORM1 transcriptional activation, while serving as a corepressor for ERBB4. It inhibits E2F1 activity by promoting E2F1-HDAC1 complex formation and suppressing E2F1 acetylation, potentially compensating for RB1 loss to prevent E2F1-mediated apoptosis. As a key regulator of CDKN1A/p21(CIP1), KAP1 exhibits E3 SUMO ligase activity toward itself (via its PHD zinc finger) and IRF7 (inhibiting its transactivation), and ubiquitinates p53/TP53 for proteasomal degradation (enhanced by MAGEC2/MAGEA2). It facilitates nuclear localization of KOX1, ZNF268, and ZNF300 transcription factors, and collaborates with ZFP90 isoform 2 to maintain FOXP3's repressor activity and Treg suppressive function. In colorectal cancer cells, KAP1 likely forms a corepressor complex enabling KRAS-driven promoter hypermethylation and TSG silencing. In ESCs, it cooperates with SETDB1 to establish H3K9me3 and silence endogenous/retroviral elements independently of DNA methylation (by similar). The SETDB1-TRIM28-ZNF274 complex may recruit ATRX to zinc-finger gene exons to maintain H3K9me3 at transcriptionally active regions. During herpesvirus 8 infection, KAP1 critically suppresses lytic gene expression early in primary infection via interaction with viral LANA1 protein.
