TGF β Receptor I (TGFBR1) is a transmembrane serine/threonine kinase that forms a non-promiscuous receptor complex with TGF-beta type II serine/threonine kinase receptor (TGFBR2) to bind TGF-beta cytokines TGFB1, TGFB2, and TGFB3. It transduces signals from these cytokines from the cell surface to the cytoplasm, regulating diverse physiological and pathological processes such as cell cycle arrest in epithelial and hematopoietic cells, mesenchymal cell proliferation/differentiation, wound healing, extracellular matrix production, immunosuppression, and carcinogenesis. The receptor complex, composed of 2 TGFBR1 and 2 TGFBR2 molecules symmetrically bound to the cytokine dimer, enables TGFBR2 (constitutively active) to phosphorylate and activate TGFBR1. Activated TGFBR1 phosphorylates SMAD2, which dissociates from the receptor, binds SMAD4, and translocates to the nucleus to modulate transcription of TGF-beta-regulated genes, forming the canonical SMAD-dependent signaling cascade. TGFBR1 also mediates non-canonical SMAD-independent pathways, e.g., inducing TRAF6 autoubiquitination to activate MAP3K7 for apoptosis or phosphorylating PARD6A to regulate epithelial-mesenchymal transition.
