Transcription Factor E3 Antibody (YA5514) is a Mouse-derived and non-conjugated IgG1 monoclonal antibody, targeting to Transcription Factor E3.

Background

TFE3 is a transcription factor that acts as a master regulator of lysosomal biogenesis and immune response (PubMed: 2338243, 24448649, 29146937, 30733432, 31672913, 37079666). It specifically recognizes and binds E-box sequences (5'-CANNTG-3'), with efficient DNA-binding requiring dimerization with itself or other MiT/TFE family members such as TFEB or MITF (PubMed: 24448649). TFE3 is involved in the cellular response to amino acid availability by acting downstream of MTOR: nutrient-rich conditions promote MTOR-mediated TFE3 phosphorylation and inactivation (PubMed: 24448649, 31672913, 36608670), while starvation or lysosomal stress inhibits MTOR, leading to TFE3 dephosphorylation and transcriptional activation (PubMed: 24448649, 31672913, 36608670). TFE3 specifically binds the CLEAR-box sequence (5'-GTCACGTGAC-3') in the regulatory regions of lysosomal genes, driving their expression and playing a central role in lysosomal gene regulation (PubMed: 24448649). It maintains embryonic stem cell pluripotency by promoting genes like ESRRB, with mTOR-dependent cytoplasmic retention and inactivation facilitating pluripotency exit (By similarity). TFE3 also inhibits neuronal progenitor differentiation (By similarity) and positively regulates adipose tissue browning by activating target genes, while mTOR-dependent phosphorylation promotes its cytoplasmic retention and suppresses browning (By similarity). In T-cells, TFE3 cooperates with TFEB to activate CD40L expression, contributing to T-cell-dependent antibody responses and thymus-dependent humoral immunity (By similarity).
