TATA Box Binding Protein Antibody (YA3260) is a rabbit-derived non-conjugated IgG monoclonal antibody (Clone NO.: YA3260), targeting TATA Box Binding Protein. It can be used as a loading control antibody.

Background

TATA Box Binding Protein (TBP) is a core component of the TFIID basal transcription factor complex, playing a pivotal role in the initiation of RNA polymerase II (Pol II)-dependent transcription. TBP recognizes and binds promoters with or without a TATA box, facilitating the assembly of the pre-initiation complex (PIC). The TFIID complex comprises TBP and TBP-associated factors (TAFs), including TAF1 to TAF13, and is structurally divided into three modules: TFIID-A, TFIID-B, and TFIID-C, with TBP forming the TFIID-A module alongside TAF3 and TAF5. During PIC assembly, TBP binds and bends promoter DNA. Additionally, TBP is part of a BRF2-containing transcription factor complex that regulates RNA polymerase III-mediated transcription, and it is a component of the SL1/TIF-IB transcription factor complex involved in RNA polymerase I-dependent PIC assembly. SL1 also stabilizes the binding of nucleolar transcription factor 1/UBTF to rDNA.

  • Western blot analysis of extracts from HepG2(lane 2(20ug) , Hela(lane 3(20ug) and HEK293(lane 4(20ug) using TATA Box Binding Protein Antibody (HY-P83515) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
  • Immunohistochemical analysis of paraffin-embedded Mouse spleen tissue using TATA Box Binding Protein Antibody (YA3260). The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 2 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P83515, 1/100) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
