Smad1 Antibody (YA077) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Smad1.

Background

SMAD1, a versatile transcriptional modulator, plays pivotal roles in various cellular processes, including embryonic development, cell differentiation, and tissue homeostasis. Upon binding of BMP ligands to their receptors, SMAD1 undergoes phosphorylation by activated type I BMP receptors, forming a heteromeric complex with SMAD4 that translocates into the nucleus, functioning as a transcription factor. This hetero-trimeric complex recognizes cis-regulatory elements containing Smad Binding Elements (SBEs), thus modulating the signaling network's outcome. Notably, the SMAD1/OAZ1/PSMB4 complex participates in the degradation of the CREBBP/EP300 repressor SNIP1 and positively regulates BMP4-induced expression of the odontogenic development regulator MSX1. Interactions with various partners such as SMAD4, YY1, HGS, NANOG, ZCCHC12, PEBP2-alpha subunit, CREB-binding protein (CBP), p300, SMURF1, SMURF2, USP15, HOXC8, ZNF423, ZNF521, SKOR1, LEMD3, PSMB4, OAZ1, FAM83G, ZC3H3, TMEM119, ZNF8, RANBP3L, EGR1, SMAD6, and YAP1 highlight the complex and dynamic nature of SMAD1's regulatory network, emphasizing its central role in cellular processes.

  • Western blot analysis of extracts from HEK293(lane 2(20μg), C2C12(lane 3(20μg) and NIH3T3(lane 4(20μg) using Smad1(HY-P80323) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
  • Immunocytochemistry analysis of Hela cells labeling Smad1 with Smad1 Antibody (HY-P80323)at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with Smad1 Antibody (HY-P80323) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L(HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).
  • Immunocytochemistry analysis of A431 cells labeling Smad1 with Smad1 Antibody (HY-P80323) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature.
