Background

p70 S6 kinase α is a serine/threonine-protein kinase that acts downstream of mTOR signaling in response to growth factors and nutrients, promoting cell proliferation, growth, and cell cycle progression. It regulates protein synthesis by phosphorylating EIF4B, RPS6, and EEF2K, and contributes to cell survival by suppressing the pro-apoptotic function of BAD. Under nutrient depletion, its inactive form associates with the EIF3 translation initiation complex, while mitogenic stimulation leads to mTORC1-mediated phosphorylation, dissociation from EIF3, and activation. The active kinase then phosphorylates substrates in the pre-initiation complex, including EIF2B and the cap-binding component EIF4B. It also controls translation initiation by phosphorylating PDCD4, targeting it for degradation. In response to IGF1, it activates translation elongation by inhibiting EEF2K, thereby activating EEF2. Additionally, it feedback-inhibits mTORC2 and AKT1 signaling by phosphorylating MAPKAP1/SIN1, MTOR, and RICTOR, and modulates mTORC1/mTORC2 activity via DEPTOR phosphorylation. It mediates cell survival by phosphorylating BAD (by similarity). Phosphorylation of mitochondrial URI1 releases PPP1CC, which dephosphorylates RPS6KB1 at Thr-412, potentially as a negative feedback mechanism. It induces TNF-α-mediated insulin resistance by accelerating IRS1 degradation via serine phosphorylation. In TSC1-2-deficient cells, it constitutively inhibits GSK3B. It may participate in cytoskeletal reorganization through neurabin binding (by similarity). Downstream of mTOR, it activates CAD, a pyrimidine biosynthesis enzyme. Upon mTORC1 activation, it phosphorylates EPRS, playing a key role in adipocyte fatty acid uptake and likely interferon-gamma-induced translation inhibition.
