Background

Retinoic Acid Receptor alpha (RARα) is a receptor for retinoic acid that binds as heterodimers (e.g., RXR/RAR) to retinoic acid response elements (RAREs, such as DR1-DR5 motifs 5'-AGGTCA-3') upon ligand (all-trans or 9-cis retinoic acid) binding, regulating gene expression in diverse biological processes. In the unliganded state, RXR-RAR heterodimers associate with corepressor complexes to induce histone deacetylation and transcriptional repression; ligand binding triggers corepressor dissociation and coactivator recruitment for transcriptional activation. Complex formation with histone deacetylases may inhibit RARE binding, while KLF2 supports RARE binding and activation. RARA is essential for retinoic acid-mediated germ cell development during spermatogenesis (by similar) and promotes survival of early meiotic spermatocytes (by similar). Collaborates with RARG in skeletal growth and growth plate homeostasis (by similar). With RXRA, it upregulates microRNA-10a to suppress GATA6/VCAM1 signaling in endothelial cells under shear stress, while HDAC3/5/7-associated RARα represses microRNA-10a to enhance inflammation.

  • Western blot analysis of extracts from NIH/3T3(lane2(20μg) , HEK293(lane 3(20μg) and MCF-7(lane 4(20ug) using Retinoic Acid Receptor alpha Antibody (HY-P80308) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
