RAGE Antibody is a Rabbit-derived and non-conjugated IgG polyclonal antibody, targeting to RAGE.

Background

RAGE is a cell surface pattern recognition receptor that senses diverse endogenous stress signals, including advanced glycation end products (AGEs), S100 proteins, high-mobility group box 1 protein (HMGB1), amyloid beta (APP) oligomers, nucleic acids, histones, phospholipids, and glycosaminoglycans. These ligands accumulate at inflammatory sites during the pathogenesis of various diseases such as diabetes, vascular complications, neurodegenerative disorders, and cancers, where RAGE transduces their binding into pro-inflammatory responses. Upon ligand binding, RAGE utilizes TIRAP and MYD88 as adapters to activate NF-kappa-B, leading to the induction of inflammatory cytokines IL6, IL8, and TNFalpha. By similarity, RAGE interacts with S100A12 to trigger cellular activation in endothelial cells, mononuclear phagocytes, and lymphocytes; with S100B to potentially promote myocyte apoptosis post-myocardial infarction via ERK1/2 and p53/TP53 signaling; and participates in amyloid-beta peptide (ABPP) translocation across neuronal membranes. It also collaborates with HMGB1 in endothelial albumin transcytosis through the RAGE/SRC/Caveolin-1 pathway and mediates HMGB1 loading into extracellular vesicles (EVs) for transferrin-mediated hepatocyte delivery, activating NLRP3 inflammasome-induced pyroptosis. By similarity, RAGE binds DNA to facilitate hypomethylated CpG DNA uptake, enhancing inflammatory responses. Additionally, it promotes non-professional phagocyte (NPP) phagocytosis of Saccharomyces cerevisiae spores, apoptotic cells, and other ligands like RNA and histones. By similarity, RAGE localizes to DNA double-strand breaks, interacts with the MRN repair complex to enhance MRE11 endonuclease activity, supports DNA damage repair in trophoblasts for placental development, and stabilizes the MCM2-7 complex during replication stress to ensure faithful fork progression.

  • Western blot analysis of extracts from M-Lung using RAGE Antibody. Proteins were transferred to a PVDF membrane and blocked with 5% nonfat powdered milk in PBST for 2 hour at room temperature. The primary antibody (1/2000) and Loading control antibody (GAPDH, 1/3000) was diluted with 5% nonfat powdered milk in PBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/8,000) was incubated for 45min at room temperature.
