RAB7 Antibody (YA110) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to RAB7.

Background

RAB7 is a small GTPase belonging to the Rab family, serving as a key regulator of intracellular membrane trafficking, from vesicle formation to membrane fusion. It cycles between GDP-bound (inactive) and GTP-bound (active) states, recruiting downstream effector proteins to mediate vesicle formation, movement, tethering, and fusion. In its active form, RAB7A plays a central role in endo-lysosomal trafficking, governing early-to-late endosomal maturation, microtubule-directed endosomal migration/positioning, and endosome-lysosome transport via protein interaction cascades. It is involved in growth factor signaling, nutrient transporter-mediated uptake, neurotrophin transport in neuronal axons, and lipid metabolism, while also regulating specialized trafficking pathways like melanosome maturation, pathogen-containing phagosome/vacuole maturation, and autophagosome formation. It contributes to phagosome acidification and fusion with lysosomes during pathogen clearance (e.g., S. aureus and M. tuberculosis) and collaborates with RAC1 to regulate osteoclast ruffled border formation. RAB7 controls NTRK1/TRKA endosomal trafficking and neurite outgrowth signaling, modulates EGF-EGFR lysosomal degradation, and participates in ADRB2-stimulated lipolysis via lipophagy (a cytosolic lipase-independent pathway, by similarity). It facilitates exosomal release of SDCBP, CD63, and syndecan (PubMed:22660413), mediates ACE2 vesicular trafficking and surface expression (PubMed:33147445), and may influence PRPH neuronal intermediate filament assembly (by similarity).

  • Immunocytochemistry analysis of NIH3T3 cells labeling RAB7 with RAB7 antibody (HY-P80295) at 1/100 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with RAB7 antibody (HY-P80295) at 1/100 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L(HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

  • Western blot analysis of extracts from Hela (lane 2(20μg), C2C12 (lane 3(20μg), C6 (lane 4(20μg) using CD44 (HY-P80295) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody (HY-P80295, 1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.
  • Flow cytometric analysis of 1X10^6 K562 cells labeling RAB7 Antibody (HY-P80295, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/50 dilution for an hour at 4℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L (HY-P8002) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Rabbit IgG Isotype Control (HY-P80879, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
