Rab10 Antibody (YA4117) is a mouse-derived and non-conjugated IgG1 antibody, targeting to Rab10. It can be applicated for WB, ICC/IF, ELISA assays, in the background of human, mouse.

Background

Rab 10 is a small GTPase that plays a key regulatory role in intracellular membrane trafficking, including the formation of transport vesicles and their fusion with membranes. It cycles between an inactive GDP-bound form and an active GTP-bound form, which recruits downstream effector proteins responsible for vesicle formation, movement, tethering, and fusion. Rab 10 is primarily involved in the biosynthetic transport of proteins from the Golgi to the plasma membrane, such as regulating the delivery of SLC2A4/GLUT4 glucose transporter-enriched vesicles to the plasma membrane (by similarity). It also regulates the transport of TLR4, a toll-like receptor, to the plasma membrane, potentially contributing to innate immune responses (by similarity). In neurons, Rab 10 is involved in axonogenesis by regulating vesicular membrane trafficking toward the axonal plasma membrane (by similarity). In epithelial cells, it controls transport from the Golgi to the basolateral membrane. Rab 10 may also participate in the basolateral recycling pathway and phagosome maturation (by similarity) and plays a role in endoplasmic reticulum dynamics, controlling tubulation along microtubules and tubule fusion. Together with LRRK2, RAB8A, and RILPL1, Rab 10 regulates ciliogenesis. When phosphorylated by LRRK2 at Thr-73, it binds RILPL1 and inhibits ciliogenesis. Additionally, Rab 10 participates in the export of newly synthesized proteins via a Rab8-Rab10-Rab11-dependent endosomal pathway. Under lysosomal stress, Rab 10 is targeted and stabilized by LRRK2 phosphorylation, promoting extracellular release of lysosomal content through EHBP1 and EHNP1L1 effector proteins. During microbial infection, Rab 10 facilitates endoplasmic reticulum recruitment and bacterial replication in Legionella pneumophila infection, aiding in the remodeling of the Legionella-containing vacuole (LCV) into an endoplasmic reticulum-like vacuole.
