Background

PUM2 is a sequence-specific RNA-binding protein that acts as a post-transcriptional repressor by binding the 3'-UTR of mRNA targets. It recognizes the Pumilio Response Element (PRE) 5'-UGUANAUA-3', related to the Nanos Response Element (NRE). PUM2 represses transcripts via multiple mechanisms: direct recruitment of the CCR4-POP2-NOT deadenylase complex (inducing mRNA decay/translational inhibition) or deadenylation-independent repression by enhancing miRNA accessibility. It post-transcriptionally suppresses E2F3 mRNAs via 3'-UTR binding and miRNA facilitation, participates in cytoplasmic viral sensing, and represses genes critical for genomic stability (e.g., mitosis/DNA repair/replication factors). This repression is modulated by the lncRNA NORAD, which sequesters PUM1/PUM2 via abundant binding sites. PUM2 may regulate DCUN1D3 mRNA levels, support stem cell proliferation/self-renewal, and—with PUM1—binds the MIR199A precursor to post-transcriptionally control its expression.
