PRKAA1 is the catalytic subunit of AMP-activated protein kinase (AMPK), an energy-sensing kinase central to cellular energy homeostasis. Under ATP-depleting conditions, AMPK activates catabolic pathways while inhibiting anabolic processes by phosphorylating metabolic enzymes (e.g., ACACA, ACACB, GYS1) and transcriptional regulators (e.g., CRTC2/TORC2, FOXO3, SREBF1/2). It enhances glucose uptake in muscle via SLC2A4/GLUT4 translocation, promotes autophagy (through ATG1/ULK1 and WDR45 activation), and modulates mitochondrial dynamics (via FOXO3 phosphorylation). PRKAA1 also regulates Wnt signaling (by stabilizing CTNNB1), circadian rhythms (via CRY1 destabilization), and stress-responsive transcription (through H2BS36ph). Key roles include lipid metabolism control (inactivating ACACA/ACACB/LIPE), mTORC1 inhibition (phosphorylating TSC2/RPTOR/WDR24), and lysosomal biogenesis (via TFEB/TFE3 nuclear translocation upon FNIP1 phosphorylation). Some functions are inferred by similarity, such as tau kinase activity (MAPT phosphorylation) and hepatic gluconeogenesis regulation (CRTC2 cytoplasmic sequestration). Stress-induced mitochondrial fragmentation is mediated via MTFR1L phosphorylation.
